Your modifications sound like they can be significant but there is not enough detail in what you plan to do to say for sure.
For your first modification you plan to do this could be significant if you are redesigning the game around the idea and it comes across clearly in game play.
The new look of enemies could be significant if you are making the sprites for these new enemies yourself.
If you are changing the entire sound scape of your game this would be significant.
